02 / THE SCIENCE BEHIND THE SELECTION

WHY THESE
INGREDIENTS?

A closer look at oxidative stress, glutamate signaling, mitochondrial energy, serotonin synthesis and sleep.

NMDA receptors / Calcium influx / Jaw tension

MAGNESIUM.

MDMA can increase glutamate release. Excessive NMDA receptor activation allows calcium to enter neurons, contributing to excitotoxicity. Magnesium is the natural voltage-dependent blocker of NMDA channels and also helps regulate muscle contraction, including jaw tension.

ROS / Antioxidant recycling / Drug metabolism

VITAMIN C.

MDMA metabolism and elevated body temperature are linked to increased production of reactive oxygen species (ROS). Vitamin C neutralizes free radicals and regenerates oxidized vitamin E, helping maintain antioxidant defenses against oxidative stress.

Cell membranes / Lipid peroxidation

VITAMIN E.

Oxidative stress associated with MDMA can trigger lipid peroxidation, a chain reaction that damages cell membranes. Vitamin E is a fat-soluble antioxidant that interrupts this process, while vitamin C helps regenerate it.

Mitochondria / ATP / Ion pumps

COQ10.

MDMA-related neuronal stress increases the energy needed to maintain ion balance. CoQ10 transfers electrons in the mitochondrial respiratory chain, supporting ATP production. ATP powers the pumps that help neurons restore calcium and other ion gradients.

Green tea catechins / Oxidative stress / Water balance

EGCG.

MDMA-related oxidative stress involves reactive molecules that can damage cellular proteins and membranes. EGCG, a green-tea catechin, has antioxidant activity and influences cellular oxidative-stress pathways studied in experimental models.

Mitochondrial metabolism / ALA combination

ALCAR.

MDMA-related oxidative stress can affect mitochondrial function. Acetyl-L-carnitine (ALCAR) supports mitochondrial metabolism and has been studied alongside alpha-lipoic acid for effects on oxidative-stress markers and neuronal function.

ROS / RNS / Antioxidant regeneration

ALPHA LIPOIC ACID (ALA).

MDMA exposure can increase reactive oxygen and nitrogen species (ROS/RNS). Alpha-lipoic acid and its reduced form, DHLA, participate in antioxidant recycling, including regeneration of vitamin C and glutathione. Animal studies have reported reduced markers of MDMA-related neuronal damage with ALA.

Cysteine / Glutathione / Cellular redox balance

NAC.

MDMA-related oxidative stress places demand on the glutathione antioxidant system. NAC supplies cysteine, a building block of glutathione (GSH). Glutathione-dependent enzymes help neutralize peroxides and maintain cellular redox balance.

Polyphenols / Inflammation / Oxidative signaling

RESVERATROL.

MDMA-related neuronal stress involves oxidative and inflammatory signaling. Resveratrol is a polyphenol studied for its effects on antioxidant responses, inflammatory pathways and cellular survival signaling in experimental models.

Sleep cycle / Free radicals / Post-event rest

MELATONIN.

MDMA can disrupt sleep after an event. Melatonin helps regulate the sleep–wake cycle and also acts as a free-radical scavenger, interacting with antioxidant enzyme systems involved in cellular defense.

03 / THE BIGGER PICTURE

HOW IT ALL CONNECTS.

Oxidative stress: MDMA metabolism and related cellular processes generate oxidative stress in experimental models. Antioxidant systems neutralize ROS/RNS and limit damage to lipids and proteins.

Excitotoxicity: Excess glutamate activates NMDA receptors, increases calcium influx and can trigger excitotoxic damage.

Mitochondrial energy: ATP powers neuronal ion pumps; CoQ10 participates in electron transport, while ALCAR supports mitochondrial metabolism.

Serotonin synthesis: MDMA releases serotonin and can alter tryptophan hydroxylase (TPH) activity. TPH is needed for serotonin synthesis; 5-HTP enters the pathway after this enzyme.

Metabolism and temperature: MDMA is metabolized through multiple liver enzymes, including CYP pathways. Elevated body temperature can intensify oxidative stress and other physiological strain.
